چکیده

The discovery of analytical tools for biomolecular recognition in the area of diagnostics is a research trend rich in innovative methods. Some of the main drivers found in this area are the constant need for low cost devices and biosensors, simplicity in design and operation, and time efficiency. In this thesis we present novel approaches for the development of protease activity assays. Our focus of study is the protease assay development process on two types of characterized platforms: microspheres and selfassembled monolayers (SAMs) on flat surfaces. The surface preparation and conjugation process with biological components of interest is presented along with surface plasmon resonance (SPR) and flow cytometry results. These platforms were engineered in order to develop assays for bacterial toxin activity. Bacterial toxins are comprised in part of proteases, which selectively cleave peptides bonds in proteins. In this thesis we present, protease assays from our main focus of studies, the Clostridium botulinum Neurotoxin type A Light Chain (BoNTA/LC) metalloprotease. Supplementary studies include assays performed involving the Bacillus anthracis Lethal factor metalloprotease. In the work described we have prepared active

G-protein-coupled receptors in drug discovery: nanosizing using cell-free technologies and molecular biology approaches.

Signal transduction by G-protein-coupled receptors (GPCRs) underpins a multitude of physiological processes. Ligand recognition by the receptor leads to activation of a generic molecular switch involving heterotrimeric G-proteins and guanine nucleotides. Signal transduction has been studied extensively with both cell-based systems and assays comprising isolated signaling components. Interest an...

Effect of Biomolecular Conformation on Docking Simulation: A Case Study on a Potent HIV-1 Protease Inhibitor

Human immunodeficiency virus infection / acquired immunodeficiency syndrome (HIV/AIDS) is a disease pertained to the human immune system. Given its crucial role in viral replication, HIV-1 protease (HIV-1 PR) is a prime therapeutic target in AIDS therapy. In this regard, the dynamic aspects of ligand-enzyme interactions may indicate an important role of conformational variability in HIV-1 PR in...

One of the important parts of innate immunity is complement system that occurs in three different ways; the classic, the alternative and the lectin pathway. The four pattern recognition molecules that have been identified till now are Mannose binding lectin (MBL), a component of lectin pathway, and three ficolins (ficolin1,-2 and -3) which compound to the carbohydrates of the cell surface. MBL ...
